Why is Mayzent prescribed?

Mayzent (Siponimod) is used to prevent symptom flare-ups and slow the progression of disability in adults with relapsing-remitting multiple sclerosis (MS), a disease that affects nerve function and can cause weakness, numbness, loss of muscle coordination, and problems with vision, speech, and bladder control.

What does it do?

Mayzent targets immune cells known as lymphocytes, which are involved in the inflammatory response in the central nervous system. By affecting these cells, Mayzent helps lower the frequency of relapses in multiple sclerosis.

How should I take it?

Take Mayzent exactly as your doctor prescribes. Follow all instructions on your prescription label and your condition since this can be on titration dose or patient information provided. Do not crush, break, or split the tablet—swallow it whole. Mayzent can be taken with or without food.

What if I missed a dose?

Contact your doctor for guidance if you miss a dose. If you miss one or more of your initial doses, or four or more maintenance doses, you may need to take your next dose under medical supervision in a healthcare setting.

What are the special precautions before taking Mayzent?

Inform your doctor about all the medications you are taking, your current and previous medical history.

Do not take Mayzent if:

  • You are allergic to siponimod or any of the ingredients in the tablets
  • You have a specific genetic profile (CYP2C9*3/*3 genotype)
  • You have certain heart conditions, including:
    • Second- or third-degree atrioventricular (AV) block
    • Irregular or abnormal heart rhythm (unless you have a pacemaker)
    • Recent (within the past 6 months) stroke, heart failure, heart attack, mini-stroke (TIA), unstable angina, or other serious heart issues

    Before starting Mayzent, inform your doctor if:

      You’ve never had chickenpox or received the varicella (chickenpox) vaccine—you may need to get vaccinated and wait 1 month before starting treatment.

      You have or have had any of the following conditions:

      • Diabetes
      • Liver disease
      • High blood pressure
      • Irregular or slow heartbeats
      • An eye condition called uveitis
      • Fever, or an active or long-term infection
      • Skin cancer (especially melanoma)
      • Breathing issues, such as asthma or sleep apnea
      • A weakened immune system (due to illness or certain medications)
      • A history of heart problems, heart attack, stroke, or chest pain

      Also tell your doctor if you’ve recently received a vaccine or have one scheduled.

      Mayzent may cause your heart rate to slow when you first start taking it. Because of this, your initial dose might be given in a setting where your heart rhythm can be closely monitored. If you miss one or more doses, you may need to restart the medication under medical supervision.

      Does it have any side effects?

      It is important to note that all drugs have corresponding side effects and adverse reactions. Additional information may be obtained when you ask your doctor or pharmacist.

        Common side effects may include:
      • Headache
      • Increased blood pressure
      • Abnormal liver function test results
        Serious side effects may include:
      • Seek emergency medical attention if you experience signs of an allergic reaction to Mayzent, such as difficulty breathing, hives, or swelling of the face, tongue, or throat.
      • Slow or irregular heartbeat
      • Seizures
      • Shortness of breath
      • Vision changes, including blurred vision, sensitivity to light, shadows in central vision, or unusual color perception
      • Headache, confusion, or changes in mental status
      • Mouth or throat sores or sores in the genital or anal area
      • Liver issues, such as dark urine, upper abdominal pain, or yellowing of the skin or eyes (jaundice)
      • Possible symptoms of meningitis, including chills, body aches, fever, fatigue, nausea, and vomiting

      Long-term use of Mayzent (over 18 months) may increase the risk of progressive multifocal leukoencephalopathy (PML), a rare but serious brain infection.

      Is it safe to take it with another drug?

      Some drug interactions may cause serious or life-threatening reactions when administered together. Immediately inform your doctor if you are taking Mayzent with any of the following medications:

      Some medications used to treat heart rhythm problems may cause harmful interactions with Mayzent. Your doctor may need to adjust your treatment if you are taking any of the following:

      • Amiodarone
      • Disopyramide
      • Dofetilide
      • Dronedarone
      • Flecainide
      • Ibutilide
      • Procainamide
      • Propafenone
      • Quinidine
      • Sotalol

      Tell your doctor about all the medicines you are currently using. Many drugs can interact with Siponimod, especially:

      • Heart medications
      • Medications that suppress the immune system, such as steroids, cancer treatments, and drugs used to prevent organ transplant rejection
      • Other medicines used to manage MS symptoms
      This is not a complete list, it may also interact with Siponimod. Be sure to inform your doctor about all the medications you are taking.

      How should I store it?

      Store unopened Mayzent tablets in the refrigerator. Do not freeze them. After opening, keep the tablets in the original container at room temperature, away from moisture and heat, and use within 3 months.

      What action should I take in case of overdose?

      If the patient has overdosed and experienced serious symptoms below, please call emergency services immediately and for monitoring.
      • Bradycardia
      • Blood and pulse monitoring

      Is it safe for pregnant or breastfeeding women?

      Mayzent may harm an unborn baby. Use effective birth control while taking the medication and for at least 10 days after your final dose. Let your doctor know if you are pregnant or plan to become pregnant.

      If you are pregnant while taking Mayzent, your name may be added to a pregnancy registry to help track outcomes.

      Consult your doctor before breastfeeding while on Mayzent to determine if it is safe.
