Why is Promacta prescribed?

Promacta (Eltrombopag) is prescribed for conditions that involve low platelet counts. It is used to treat chronic immune thrombocytopenia (ITP) in patients who have not responded adequately to corticosteroids, immunoglobulins, or splenectomy, helping to reduce the risk of bleeding..

Additionally, it is prescribed to increase platelet levels in individuals with chronic hepatitis C-associated thrombocytopenia, enabling them to begin and sustain interferon-based therapy. For severe aplastic anemia (SAA), Promacta is used alongside immunosuppressive treatment as a first-line therapy or for patients who have not responded to previous immunosuppressive therapy..

What does it do?

Promacta (Eltrombopag) stimulates platelet production by activating thrombopoietin receptors in the bone marrow. It is primarily used to increase platelet counts in people with conditions that cause low platelets, such as chronic immune thrombocytopenia (ITP), hepatitis C-associated thrombocytopenia, and severe aplastic anemia (SAA). However, it is not intended to normalize platelet counts but rather to maintain a safe level to reduce bleeding risks.

How should I take it?

Promacta should be taken on an empty stomach or with a meal low in calcium (=50 mg). It’s important to take it at least 2 hours before or 4 hours after any medications or foods containing polyvalent cations (such as antacids, calcium-rich foods, and mineral supplements) because they can interfere with absorption.

The initial dosage varies based on the specific condition being treated. See the guidelines below:

  • Chronic Immune Thrombocytopenia (ITP): The usual dose is 50 mg once daily for adults and children 6 years and older. For East-/Southeast-Asian ancestry or those with hepatic impairment, the starting dose is 25 mg once daily.
  • Chronic Hepatitis C-associated Thrombocytopenia: The usual dose is 25 mg once daily.
  • Severe Aplastic Anemia (SAA): Promacta is used in combination with immunosuppressive therapy.
Your doctor may adjust the dose based on platelet counts and liver function tests, ensuring it does not exceed the maximum limit of 75 mg per day.

What if I missed a dose?

If you miss a dose of Promacta, take it as soon as you remember. If your next dose is approaching, skip the missed one and continue following your usual schedule. Avoid taking extra or double doses to compensate for the missed dose.

What are the special precautions before taking Promacta?

Before taking Promacta, here are several important precautions to keep in mind:

  • Promacta may increase the risk of severe hepatotoxicity. Liver function should be monitored regularly, especially in patients with chronic hepatitis C.
  • Disclose all your medical history to your doctor, particularly if you have had:
    • Pre-cancerous condition called myelodysplastic syndrome (MDS) or blood cancer
    • Bleeding or blood clotting disorder
    • Cataracts
    • Surgery to remove your spleen
    • Liver disease
  • The dose should be carefully adjusted to avoid excessive platelet production, as high platelet counts can increase the risk of thrombosis (blood clots).
  • Promacta is not recommended for patients with myelodysplastic syndrome (MDS) due to potential risks.
  • Avoid taking Promacta with calcium-rich foods or polyvalent cation-containing medications (such as antacids and mineral supplements), as they can reduce absorption.

Does it have any side effects?

It is important to note that all drugs have corresponding side effects and adverse reactions. Additional information may be obtained when you ask your doctor or pharmacist.
    Common side effects may include:
  • Fatigue
  • Headache
  • Nausea
  • Muscle aches
  • Vomiting
    Serious side effects may include:
  • Cataracts (eye problems)
  • Difficulty breathing
  • Sudden severe headache
  • Increased risk of blood clots
  • Severe bleeding
This is not a complete list of all possible side effects, and others may occur. For more information about side effects, please consult your healthcare provider.

Is it safe to take it with another drug?

Promacta (Eltrombopag Olamine) can interact with various medications, so it's important to check for potential interactions before taking it with another drug.

Some drugs can significantly affect Promacta’s effectiveness or increase the risk of side effects. These include:

  • Antacids & Calcium Supplements
  • Blood pressure medications
  • Certain laxatives
  • Diabetes medications
  • Cancer treatment drugs
  • Methotrexate
  • Statins (Atorvastatin)
This is not a complete list of all the potential drug interactions, always consult your doctor or pharmacist before combining Promacta with other medications to ensure safety.

How should I store it?

Promacta should be stored in its original container at room temperature, ideally between 20°C and 25°C (68°F and 77°F).

What action should I take in case of overdose?

If you suspect an overdose of Promacta (Eltrombopag Olamine), it's important to act quickly and seek immediate medical attention. Contact emergency services or go to the nearest hospital. Symptoms of an overdose may include severe bleeding, dark urine, dizziness, or sudden vision changes.

Is it safe for pregnant or breastfeeding women?

Promacta (Eltrombopag) should be used during pregnancy only if the benefits outweigh the risks. There is limited human data on its effects during pregnancy, but animal studies suggest potential risks, including embryolethality and reduced fetal weight. For breastfeeding, it is unknown whether Promacta passes into human milk, but it has been detected in animal studies. The manufacturer recommends avoiding breastfeeding while taking Promacta.

If you are pregnant, planning to become pregnant, or breastfeeding, consult your healthcare provider for guidance on whether Promacta is appropriate for you.

Canadian Prescription Drug Store dispenses medications from our fully licensed and independently verified pharmacy partners around the world from Canada, England, Australia, New Zealand, India, and Mauritius.

Legal disclaimer: The FDA, due to the current state of their regulations, has taken the position that virtually all shipments of prescription drugs imported from a Canadian online pharmacy by a U.S. consumer will violate the law. However, many legal experts disagree with the FDA's position in this regard stating that such importation is permitted under certain federal and state laws. Several State governments and consumer rights groups actively promote the importation of medications. The FDA itself provides the public information on how to import medications safely. Furthermore, there is no record of any US individual being prosecuted by the FDA for importing non-controlled prescription drugs for their personal use.
