Why is Afinitor prescribed?

Afinitor (Everolimus) is a cancer medication that disrupts the growth of cancer cells and helps slow their spread throughout the body. It is used to treat specific types of kidney cancer, breast cancer, and brain tumors. Afinitor is also prescribed for certain advanced or progressive tumors of the stomach, intestines, or pancreas.

What does it do?

Afinitor is under drug class known as kinase inhibitor that works by stopping or halting the growth of cancer or tumor cells.

How should I take it?

Take this medication orally as prescribed by your doctor, typically 10mg once a day. You can take it with or without food, but it is important to be consistent and take it the same way with each dose. Swallow the tablet whole with a glass of water. Do not crush or chew the tablets.

What if I missed a dose?

If you miss a dose, take it as soon as you remember. If more than 6 hours have passed since the missed dose, skip it and take your next dose at the usual time. Do not take two doses to make up for the missed one.

What are the special precautions before taking Afinitor?

Afinitor can make you feel generally unwell, which is common as chemotherapy affects both cancer and healthy cells. Report any side effects to your healthcare provider. Continue treatment unless your doctor advises otherwise. Regular check-ups and tests may be needed to monitor for side effects.

Before undergoing surgery, consult your healthcare provider, as Afinitor can increase the risk of poor wound healing. You will need to stop taking the medication one week before surgery and wait at least two weeks after surgery before resuming it. Ensure the surgical site has healed properly before restarting the medication.

This medication may increase blood sugar levels. If you have diabetes, ask your doctor if you need any changes to your diet or medication and can cause ovarian failure in women and reduced sperm count in men, which may affect fertility. If you have concerns about fertility, talk to your healthcare provider.

If you've had a kidney transplant, inform your doctor immediately if your incision site becomes red, warm, painful, or shows signs of swelling, blood, fluid, or pus. also avoid sun exposure, and if you must be outside, wear protective clothing and sunscreen. Do not use tanning beds or sun lamps.

Does it have any side effects?

Seek emergency medical attention if you experience signs of an allergic reaction to Afinitor, such as hives; chest pain, difficulty breathing; or swelling of the face, lips, tongue, or throat. You may be at higher risk for these symptoms if you're also taking an "ACE inhibitor" for heart or blood pressure issues.

Your cancer treatment may be delayed or stopped permanently if you experience certain side effects.
    Common side effects may include:
  • Fever, cough, infections, weakness, or fatigue
  • Mouth sores
  • Nausea, loss of appetite
  • Swelling anywhere in your body
  • Rash
  • Missed periods
  • Headache
  • High blood sugar
  • Contact your doctor immediately if you have the following conditions:

  • A wound that won't heal or a surgical incision that is red, warm, swollen, painful, bleeding, or oozing pus
  • Blisters or ulcers in your mouth, trouble swallowing, swollen or red gums
  • Lung issues, including a new or worsening cough, chest pain, wheezing, or shortness of breath
  • Signs of infection, such as fever, fatigue, chills, joint pain, or a skin rash
  • Kidney problems, including little or no urination or swelling in your feet or ankles
  • Liver issues, such as nausea, loss of appetite, clay-colored stools, upper right stomach pain, dark urine, or jaundice (yellowing of the skin or eyes)
  • Low blood cell counts, with flu-like symptoms, skin sores, unusual bleeding, easy bruising, pale skin, cold hands and feet, or lightheadedness
  • This list is not exhaustive, and other side effects may occur. Contact your doctor for medical advice regarding any side effects.

    Is it safe to take it with another drug?

    Inform your doctor about all the medications you are currently taking, as many drugs can interact with Afinitor, especially:
  • Antibiotics or antifungal medications
  • Heart or blood pressure medications
  • Medicines for treating hepatitis C or HIV/AIDS
  • Seizure medications
  • St. John's Wort
  • Tuberculosis treatments
  • Drugs that suppress the immune system, including cancer treatments, steroids, and medications for organ transplant rejection

    How should I store it?

    Store the medication in its original container at room temperature, protected from light and moisture. Do not store it in the bathroom. Keep all medications out of reach of children and pets.

    What action should I take in case of overdose?

    If you believe you have taken an overdose of this medication, immediately contact your provider or go to the emergency room.

    Is it safe for pregnant or breastfeeding women?

    If you are pregnant or planning to become pregnant, inform your doctor, as you should not get pregnant while using everolimus. This medication can harm an unborn baby. Men should also discuss birth control methods with their doctor during treatment and for 4 weeks after stopping the medication. If you or your partner becomes pregnant, contact your doctor immediately to discuss the risks and benefits of continuing the medication.

    Since everolimus can be absorbed through the skin and lungs and may harm an unborn baby, pregnant women or those who may become pregnant should avoid handling the medication or inhaling tablet dust.

    It is unclear whether everolimus passes into breast milk. Due to potential risks to the infant, breastfeeding is not recommended during treatment and for 2 weeks after the last dose. Consult your doctor before breastfeeding.
